چکیده

Escherichia coli B contains a cytidine diphosphate reductase system which utilizes reduced triphosphopyridine nucleotide to effect the reduction of cytidine diphosphate to deoxycytidine diphosphate (1, 2). Two components, thioredoxin and thioredoxin reductase, have been identified as oxidation-reduction carriers between TPNH and cytidine diphosphate. Thioredoxin is a low molecular weight protein which contains two sulfhydryl groups (1). Oxidized thioredoxin, containing a disulfide group (SZ), is reduced by the TPNH-specific thioredoxin reduct.ase according to the following equat.ion.

The Stereospecificity of the Hill Reaction with Triphosphopyridine Nucleotide.

In 1957, San Pietro and Lang (1) observed that isolated spinach chloroplasts cause a light-stimulated exchange of the hydrogen of water with the hydrogen at the p position of the nicotinamide ring of oxidized TPN and DPN. Their results showed that there are enzymes in chloroplasts which cause activation of both steric positions of the reduced pyridine nucleotides. Since these early studies, it ...

Enzymatic Synthesis of Deoxyribonucleotides. Iv. Isolation and Characterization of Thioredoxin, the Hydrogen Donor from Escherichia Coli B.

The reductive formation of deoxycytidine diphosphate from cytidine diphosphate with crude extracts of Escherichia coli B required the presence of reduced triphosphopyridine nucleotide (1). On purification of the GDP-reductase system, an enzyme fraction (Fraction B) was obtained, which was no longer active with TPNH but, instead, showed an absolute requirement for reduced lipoate (2, 3). Several...

The Human Thioredoxin System: Modifications and Clinical Applications

The thioredoxin system, comprising thioredoxin (Trx), thioredoxin reductase (TrxR) and NADPH, is one of the major cellular antioxidant systems, implicated in a large and growing number of biological functions. Trx acts as an oxidoreductase via a highly conserved dithiol/disulfide motif located in the active site ( Trp-Cys-Gly-Pro- Cys-Lys-). Different factors are involved in the regulation of T...

Replacement of threonine-55 with glycine decreases the reduction rate of OsTrx20 by glutathione

Thioredoxins (Trxs) are small ubiquitous oxidoreductase proteins with two redox-active Cys residues in a conserved active site (WCG/PPC) that regulate numerous target proteins via thiol/disulfide exchanges in the cells of prokaryotes and eukaryotes. The isoforms OsTrx23 with a typical active site (WCGPC) and OsTrx20 with an atypical active site (WCTPC) are two  Trx h- type isoforms in rice that ...
